Koala cathelicidin PhciCath5 has antimicrobial activity, including against Chlamydia pecorum

چکیده

Devastating fires in Australia over 2019–20 decimated native fauna and flora, including koalas. The resulting population bottleneck, combined with significant loss of habitat, increases the vulnerability remaining koala populations to threats which include disease. Chlamydia is one disease causes morbidity mortality predominant pathogenic species, pecorum , severe ocular, urogenital reproductive tract In marsupials, koala, gene expansions an antimicrobial peptide family known as cathelicidins have enabled protection immunologically naïve pouch young during early development. We propose that are active against other bacteria fungi. Here we describe ten cathelicidins, five contained full length coding sequences were widely expressed tissues throughout body. Focusing on these five, investigate their activity two C . isolates from distinct serovars; MarsBar IPTaLE, well One cathelicidin, PhciCath5, inactivated IPTaLE elementary bodies significantly reduced number inclusions compared control (p<0.0001). Despite evidence cathelicidin expression within be infected by natural PhciCath5 concentrations may inadequate vivo prevent or infections also displayed fungi Gram negative positive bacteria, methicillin-resistant Staphylococcus aureus (MRSA). Electrostatic interactions likely drive adherence pathogen cell membrane, followed membrane permeabilisation leading death. Activity E coli was presence 10% serum 20% whole blood. Future modification enhance activity, serum/blood, provide a novel solution infection koalas species.

Activity of cathelicidin peptides against Chlamydia spp.

The in vitro activity of six cathelicidin peptides against 25 strains of Chlamydia was investigated. SMAP-29 proved to be the most active peptide, reducing the inclusion numbers of all 10 strains of Chlamydia trachomatis tested by > or =50% at 10 microg/ml. This peptide was also active against C. pneumoniae and C. felis.

Activity of Cathelicidin Peptides against Simkania negevensis

The in vitro activity of six cathelicidin peptides against the reference strain Z of Simkania negevensis was investigated. Five peptides-PG-1, Bac7, SMAP-29, BMAP-27, and BMAP-28-proved to be active at very low concentrations (1 to 0.1 μg/mL), while LL-37 cathelicidin was ineffective even at a concentration of 100 μg/mL. In comparison to chlamydiae, S. negevensis proved to be more susceptible t...

Molecular characterisation of the Chlamydia pecorum plasmid from porcine, ovine, bovine, and koala strains indicates plasmid-strain co-evolution

Background. Highly stable, evolutionarily conserved, small, non-integrative plasmids are commonly found in members of the Chlamydiaceae and, in some species, these plasmids have been strongly linked to virulence. To date, evidence for such a plasmid in Chlamydia pecorum has been ambiguous. In a recent comparative genomic study of porcine, ovine, bovine, and koala C. pecorum isolates, we identif...

Preliminary Characterisation of Tumor Necrosis Factor Alpha and Interleukin-10 Responses to Chlamydia pecorum Infection in the Koala (Phascolarctos cinereus)

Debilitating infectious diseases caused by Chlamydia are major contributors to the decline of Australia's iconic native marsupial species, the koala (Phascolarctos cinereus). An understanding of koala chlamydial disease pathogenesis and the development of effective strategies to control infections continue to be hindered by an almost complete lack of species-specific immunological reagents. The...

The Human Cathelicidin LL-37 Has Antiviral Activity against Respiratory Syncytial Virus

Respiratory syncytial virus is a leading cause of lower respiratory tract illness among infants, the elderly and immunocompromised individuals. Currently, there is no effective vaccine or disease modifying treatment available and novel interventions are urgently required. Cathelicidins are cationic host defence peptides expressed in the inflamed lung, with key roles in innate host defence again...
